Genefrei® Spleen Targeted Transfection Kit

Genefrei® spleen targeted transfection kit is a ready-to-use kit developed by Longuide Limited Lab specially for spleen-targeting RNA transfection in mice. In vivo transfection of mice can be performed only by mixing the solution in the kit with RNA, the kit possesses advantages including simple operation, low toxicity and high targeting capability. The Genefrei® spleen targeted transfection kit can accelerate the development of RNA drugs for treatment of splenic diseases and other immunotherapies.

The figure on the left shows in vivo transfection of mice using the Genefrei® spleen targeted transfection kit encapsulating Fluc mRNA and Fluc circRNA. The kit has good transfection efficiency and spleen-targeting capability for diverse nucleic acids.

We tested the transfection efficiency of Genefrei® spleen targeted transfection kit via intravenous injection after being stored at 4℃ for different time and mixed with Fluc mRNA. There was no significant change in the transfection ability and targeting capability of the Genefrei® spleen targeted transfection kit over a period of 60 days, indicating its good stability.

Blood biochemical test results show that the ALT, AST, and Crea levels increased slightly but still within the normal range 24 h post administration of 300 μg mRNA per mouse. The biochemical index returned to normal 96 h after the treatment, besides, liver and kidney function of the treatment group is normal, indicating high safety profile of the Genefrei® spleen targeted transfection kits. High safety profiles of the kits endows it with significant advantage in studies where high-dose administration is needed, such as gene editing.

The figure on the left shows that mice were given different doses of Fluc mRNA loaded with Genefrei® spleen targeted transfection kit by IV injection. The transfection efficiency in mice was significantly dose-dependent, and the bioluminescence signal increased with the increase of dose, indicating the kit holds promise in application in dose-diversified administration regimen.
